Một túi tuyến nhỏ dưới bụng một con hươu không sừng. Những hạt nâu sẫm, trong đó muscone chỉ chiếm khoảng 0,5 đến 2 phần trăm. Và một chữ, musk, nay đứng trên nhãn rất nhiều chai nước hoa không còn chứa gì từ con hươu ấy.
Một túi nhỏ và một chữ lớn
Hươu xạ là loài hươu nhỏ, không có sừng, sống trong rừng núi châu Á, từ Siberia qua dãy Himalaya xuống miền Nam Trung Quốc. Con đực mang một túi tuyến dưới bụng. Dịch tiết trong túi khô lại thành những hạt màu nâu sẫm, trong thương mại gọi là musk grains. Đó là xạ hương hươu xạ, nguyên liệu đã đặt tên cho cả một họ mùi.
Mùi của những hạt này được ghi lại là ấm, có chất phấn, gần với mùi da, thoáng nét động vật, kèm một cạnh hoa khô và mật ong. Nó toả rất xa dù chỉ một lượng nhỏ, và bám rất lâu. Trong nước hoa cổ điển, xạ hương được dùng dưới dạng cồn ngâm: hạt khô ngâm trong ethanol nhiều tháng, rồi lọc lấy phần dịch.
Có một chi tiết địa danh trong lịch sử thương mại của nó. Tên gọi Tonkin musk, hay Tonquin musk, lấy từ Tonkin, cái tên người châu Âu thời thuộc địa dùng cho miền Bắc Việt Nam. Xạ hương đi qua ngả Vân Nam và Tonkin từng được xếp hạng cao trong thương mại thế kỷ 19. Cái tên đánh dấu một con đường buôn bán, không phải một loài hươu riêng.
Ngày nay chữ musk trên một chai nước hoa gần như luôn chỉ một phân tử tổng hợp. Khoảng cách giữa con hươu và cái nhãn là một thế kỷ hoá học, và một hệ thống luật về buôn bán động vật hoang dã.
Hươu xạ và CITES
Túi xạ nằm trong cơ thể con hươu, và trong lịch sử, lấy túi đồng nghĩa với giết con vật. Khi giá xạ hương cao, áp lực săn bắt lên các quần thể hươu xạ hoang dã tăng theo.
Công ước CITES hiện xếp toàn bộ chi Moschus vào Phụ lục II, nghĩa là buôn bán quốc tế cần giấy phép và phải chứng minh không gây hại cho sự tồn tại của loài. Các quần thể ở Afghanistan, Bhutan, Ấn Độ, Myanmar, Nepal và Pakistan nằm ở Phụ lục I, mức kiểm soát chặt nhất, gần như cấm buôn bán vì mục đích thương mại.
Trung Quốc nuôi hươu xạ từ năm 1958 và phát triển cách lấy xạ từ con vật còn sống, chủ yếu cho y học cổ truyền. Trong thương mại hương liệu quốc tế, xạ hương hươu gần như vắng mặt từ cuối thế kỷ 20. Tại Việt Nam, buôn bán xạ hương thật chịu sự điều chỉnh của luật về động vật hoang dã và các quy định thực thi CITES.
Như vậy, chữ xạ hương trên một chai nước hoa bán lẻ hôm nay gần như chắc chắn không đến từ con hươu. Điều đáng hỏi hơn là nó đến từ phân tử nào, và phân tử ấy thuộc thế hệ nào.
Muscone và musk vòng lớn
Phân tử mang mùi chính của xạ hương hươu là muscone. Heinrich Walbaum tách được nó năm 1906. Hai mươi năm sau, năm 1926, Leopold Ružička chứng minh muscone là một ketone có vòng mười lăm nguyên tử carbon. Kết quả ấy thay đổi cách giới hoá học nhìn những vòng carbon lớn. Ružička nhận giải Nobel Hoá học năm 1939 cho các công trình về polymethylene và terpene.
Cùng năm 1926, Ružička xác định cấu trúc civetone từ xạ cầy hương, cũng là một ketone vòng lớn. Hai con vật khác nhau, hai tuyến khác nhau, nhưng cùng một kiểu hình học phân tử. Từ đó mở ra nhóm xạ hương vòng lớn: ketone, lactone và diester có vòng từ mười lăm nguyên tử trở lên.
Cyclopentadecanone, bán dưới tên Exaltone, là muscone bớt một nhóm methyl. Ružička làm ra nó trong thời gian hợp tác với công ty ở Geneva sau này trở thành Firmenich. Mùi được mô tả là xạ khô, sạch, có chất sáp và phấn, ít nét động vật hơn muscone. Ethylene brassylate là một diester vòng lớn, làm từ axit brassylic và ethylene glycol; axit brassylic có thể lấy từ axit erucic trong dầu hạt cải, nên nguyên liệu đầu có gốc thực vật. Giá thấp và độ bền cao đưa nó vào nhóm musk vòng lớn được dùng nhiều nhất về khối lượng.
Muscone tự nhiên là đồng phân (R). Muscone thương mại là hàng tổng hợp, ở dạng racemic hoặc giàu một đồng phân. Nó đắt hơn phần lớn các musk tổng hợp khác, nên chủ yếu nằm trong nước hoa cao cấp. Theo chỉ mục IFRA 51, muscone không có tiêu chuẩn hạn chế riêng.
Musk nitro và musk đa vòng: galaxolide, tonalide
Xạ hương tổng hợp đầu tiên không đến từ phòng thí nghiệm nước hoa mà từ nghiên cứu thuốc nổ. Năm 1888, Albert Baur thu được một hợp chất nitro có mùi xạ khi làm việc với các chất họ TNT. Musk ketone, được cấp bằng sáng chế năm 1894, trở thành nền của nhiều nước hoa cổ điển. Musk ambrette, musk xylene và các musk nitro khác nối theo.
Thế hệ nitro rẻ và bền, nhưng từ thập niên 1980 dữ liệu về tích luỹ sinh học và phản ứng với ánh sáng trên da dẫn tới các lệnh cấm lần lượt. IFRA cấm musk ambrette năm 2006, musk moskene và musk tibetene năm 2008, musk xylene năm 2009. Musk ketone vẫn được phép theo một tiêu chuẩn đặc tả, và tại EU nằm trong Phụ lục III với mức tối đa 1,4 phần trăm trong nước hoa.
Thế hệ tiếp theo là musk đa vòng. Galaxolide do các nhà hoá học của IFF tìm ra năm 1965 và nhanh chóng trở thành musk tổng hợp được dùng nhiều nhất thế giới. Nó rẻ, bền trong môi trường kiềm, mùi sạch và ngọt với nét hoa và gỗ, gần với mùi vải vừa giặt. Chính những đặc điểm đó đưa nó vào xà phòng, nước giặt và nước xả vải. Tonalide, một musk đa vòng cùng thời, thường đi cùng galaxolide; hai phân tử này từng chiếm phần lớn khối lượng musk tổng hợp toàn cầu.
Từ thập niên 1990, galaxolide được phát hiện trong nước thải, bùn và sinh vật thuỷ sinh. Nhiều đánh giá môi trường ở châu Âu và Bắc Mỹ theo sau, và ngành hương liệu dần chuyển về phía musk vòng lớn và musk alicyclic dễ phân huỷ hơn. Quy định EU 2023/1545 đưa hexamethylindanopyran, tên hoá học của galaxolide trong danh sách, vào danh sách chất gây dị ứng phải ghi trên nhãn khi vượt 0,001 phần trăm trong sản phẩm lưu trên da. Tonalide có giới hạn riêng trong Phụ lục III của EU, 2,5 phần trăm trong nước hoa.
Hạt ambrette: musk từ thực vật
Một số thực vật cũng tạo ra phân tử vòng lớn mang mùi xạ. Cây vông vang, Abelmoschus moschatus, thuộc họ Cẩm quỳ, cùng họ với cây bông và dâm bụt. Hạt của nó được thu hoạch, sấy khô rồi chưng cất hơi nước để lấy tinh dầu ambrette. Nguồn chính là Ấn Độ, Madagascar và Indonesia.
Thành phần lớn nhất của tinh dầu hạt ambrette không phải là chất mang mùi xạ. Farnesyl acetate chiếm khoảng 30 đến 65 phần trăm, farnesol từ 3 đến 39 phần trăm. Phân tử tạo nét xạ là ambrettolide, một lactone vòng mười bảy nguyên tử, chiếm khoảng 7,6 đến 14,7 phần trăm. Một cấu trúc hiếm gặp trong tinh dầu thương mại, nằm trong một hạt nhỏ.
Tỷ lệ farnesol và farnesyl acetate cao khiến dầu ambrette dễ bị oxi hoá, nên cần chai tối màu, đậy kín và giữ lạnh. Ngoài tinh dầu còn có dạng chiết bằng dung môi và bằng CO2 từ hạt, mỗi dạng cho một hồ sơ mùi khác. Rễ bạch chỉ châu Âu, angelica, là một nguồn thực vật khác, chứa pentadecanolide, cũng là một lactone vòng lớn.
Các nguyên liệu này đắt và dùng ở lượng nhỏ. Chúng không thay thế được khối lượng musk tổng hợp trong nước giặt, nhưng cho nước hoa một nét xạ có gốc thực vật, ấm và gần với xạ động vật, khác với độ sạch phẳng của musk đa vòng.
White musk trên da
Musk là nốt đáy chậm nhất trong một công thức. Phân tử lớn, khối lượng phân tử quanh 240 đến 270, bay hơi rất chậm, nên thường là thứ còn lại trên da sau nhiều giờ. White musk là cách gọi phổ biến cho kiểu xạ sạch, không nét động vật, dựng từ các phân tử musk tổng hợp như galaxolide, habanolide, ethylene brassylate hay helvetolide. Habanolide được mô tả như vải lanh vừa giặt, có chút kim loại. Helvetolide, do Firmenich đưa ra năm 1990, là musk alicyclic thương mại đầu tiên, có nét quả lê.
Một đặc điểm của musk là không phải ai cũng ngửi thấy. Nhiều người mất khứu giác đặc hiệu với một số phân tử musk nhất định: phân tử ấy có trên da, nhưng người đó không nhận ra. Vì vậy các công thức thường dùng nhiều loại musk cùng lúc, mỗi loại lấp một khoảng trống cho người không ngửi được loại kia. Hiện tượng này được bàn kỹ hơn trong bài một mùi mà một nửa căn phòng không thấy.
Mỗi thế hệ musk mang một kiểu sạch và ấm riêng. Muscone tròn, sữa, có chút da. Musk nitro ngọt, hơi bụi. Musk đa vòng ngọt, hoa, gắn với mùi quần áo. Musk vòng lớn mềm, phấn. Musk alicyclic sáng, có nét trái cây. Ambrette ấm, gần với xạ động vật hơn cả. Đọc một nốt xạ trên que thử là đọc xem nó thuộc tầng lịch sử nào.
Nhãn Xạ Hương Trắng của Lê Mai thuộc về dòng này: một nốt musk sạch dựng bằng phân tử, không có nguyên liệu từ hươu xạ. Việc chuyển từ con vật sang phân tử không chỉ là câu chuyện đạo đức. Hoá học của Ružička và những người sau ông đã làm cho sự chuyển đổi đó khả thi, từng vòng một.

A small gland pouch beneath the belly of an antlerless deer. Dark brown grains in which muscone makes up only about 0.5 to 2 percent. And one word, musk, which now sits on the labels of a great many perfumes that contain nothing from that deer.
A Small Pouch and a Large Word
The musk deer is a small deer without antlers that lives in the mountain forests of Asia, from Siberia across the Himalaya to southern China. The male carries a gland pouch under its belly. The secretion inside dries into dark brown grains, known in trade as musk grains. This is deer musk, the material that gave its name to an entire family of scent.
The grains are described as warm and powdery, close to the smell of skin, faintly animal, with an edge of dried flowers and honey. They diffuse widely in tiny amounts and last a very long time. In classical perfumery musk was used as a tincture: dried grains steeped in ethanol for months, then filtered.
Its trade history carries a place name. Tonkin musk, or Tonquin musk, takes its name from Tonkin, the colonial European name for northern Vietnam. Musk that travelled through Yunnan and Tonkin was graded highly in nineteenth century trade. The name marks a trade route, not a separate species of deer.
Today the word musk on a perfume bottle almost always refers to a synthetic molecule. Between the deer and the label lie a century of chemistry and a body of law on the trade in wild animals.
The Musk Deer and CITES
The musk pod sits inside the body of the deer, and historically taking the pod meant killing the animal. When the price of musk was high, hunting pressure on wild musk deer populations rose with it.
CITES currently lists the entire genus Moschus in Appendix II, which means international trade requires permits and evidence that it does not harm the survival of the species. The populations of Afghanistan, Bhutan, India, Myanmar, Nepal and Pakistan are in Appendix I, the strictest level, which all but excludes commercial trade.
China has farmed musk deer since 1958 and developed ways to collect musk from live animals, mainly for traditional medicine. In the international fragrance trade deer musk has been almost absent since the late twentieth century. In Vietnam, trade in natural musk falls under wildlife law and the rules that implement CITES.
So the word musk on a retail perfume today almost certainly does not come from the deer. The better question is which molecule it comes from, and which generation that molecule belongs to.
Muscone and the Macrocyclic Musks
The main odorant of deer musk is muscone. Heinrich Walbaum isolated it in 1906. Twenty years later, in 1926, Leopold Ružička showed that muscone is a ketone with a ring of fifteen carbon atoms. The result changed how chemists thought about large carbon rings. Ružička received the 1939 Nobel Prize in Chemistry for work on polymethylenes and terpenes.
In the same year Ružička determined the structure of civetone from civet, also a large ring ketone. Two different animals, two different glands, one molecular geometry. This opened the family of macrocyclic musks: ketones, lactones and diesters with rings of fifteen atoms or more.
Cyclopentadecanone, sold as Exaltone, is muscone without one methyl group. Ružička made it while working with the Geneva company that later became Firmenich. Its smell is described as a dry, clean musk, waxy and powdery, with less animal character than muscone. Ethylene brassylate is a macrocyclic diester made from brassylic acid and ethylene glycol; brassylic acid can be obtained from erucic acid in rapeseed oil, so the starting material is plant derived. Its low cost and high stability have made it one of the highest volume macrocyclic musks.
Natural muscone is the (R) enantiomer. Commercial muscone is synthetic, either racemic or enriched in one enantiomer. It costs more than most synthetic musks and is found mainly in fine fragrance. In the IFRA 51 index muscone has no dedicated restricting standard.
Nitro and Polycyclic Musks: Galaxolide, Tonalide
The first synthetic musk came not from a perfume laboratory but from research on explosives. In 1888 Albert Baur obtained a musky nitro compound while working with TNT type substances. Musk ketone, patented in 1894, became the base of many classical perfumes. Musk ambrette, musk xylene and other nitro musks followed.
The nitro generation was cheap and stable, but from the 1980s data on bioaccumulation and on light reactions on skin led to a series of bans. IFRA prohibited musk ambrette in 2006, musk moskene and musk tibetene in 2008, and musk xylene in 2009. Musk ketone remains permitted under a specification standard, and in the EU it sits in Annex III with a maximum of 1.4 percent in fine fragrance.
The next generation was the polycyclic musks. Galaxolide was discovered by IFF chemists in 1965 and soon became the most used synthetic musk in the world. It is cheap, stable in alkaline media, and smells clean and sweet with floral and woody touches, close to freshly laundered cloth. Those qualities carried it into soap, detergent and fabric softener. Tonalide, a polycyclic musk of the same period, often travels with galaxolide; the two once accounted for most of the world's synthetic musk volume.
From the 1990s galaxolide was detected in wastewater, sludge and aquatic organisms. Environmental assessments in Europe and North America followed, and the industry gradually shifted toward macrocyclic and more degradable alicyclic musks. EU Regulation 2023/1545 adds hexamethylindanopyran, the chemical name under which galaxolide appears on the list, to the allergens that must be named on the label above 0.001 percent in leave on products. Tonalide has its own limits in EU Annex III, 2.5 percent in fine fragrance.
Ambrette Seed: Musk from a Plant
Some plants also produce large ring molecules with a musky smell. Abelmoschus moschatus belongs to the mallow family, related to cotton and hibiscus. Its seeds are harvested, dried and steam distilled to give ambrette seed oil. The main producers are India, Madagascar and Indonesia.
The largest part of ambrette seed oil is not what carries the musk. Farnesyl acetate makes up about 30 to 65 percent and farnesol 3 to 39 percent. The molecule behind the musky note is ambrettolide, a lactone with a seventeen membered ring, at about 7.6 to 14.7 percent. An uncommon structure among commercial essential oils, held inside a small seed.
The high share of farnesol and farnesyl acetate makes ambrette oil prone to oxidation, so it needs a dark bottle, a tight seal and cold storage. Besides the oil there are an absolute and a CO2 extract of the seed, each with a different scent profile. Angelica root is another plant source, containing pentadecanolide, also a macrocyclic lactone.
These materials are costly and used in small amounts. They cannot replace the tonnage of synthetic musk in laundry products, but they give perfume a musk of plant origin, warm and close to animal musk, unlike the flat cleanness of the polycyclics.
White Musk on Skin
Musk is the slowest base note in a formula. The molecules are large, with molecular weights around 240 to 270, and evaporate very slowly, so they are often what remains on skin after many hours. White musk is the common name for a clean musk without animal character, built from synthetic musk molecules such as galaxolide, habanolide, ethylene brassylate or helvetolide. Habanolide is described as freshly washed linen with a slightly metallic edge. Helvetolide, introduced by Firmenich in 1990, was the first commercial alicyclic musk, with a pear like nuance.
One feature of musk is that not everyone can smell it. Many people have a specific anosmia to certain musk molecules: the molecule is on the skin, but that person does not perceive it. For this reason formulas often use several musks at once, each filling a gap for people who miss another. The phenomenon is discussed further in a smell half the room cannot detect.
Each generation of musk carries its own kind of clean and warm. Muscone is round and milky with a trace of skin. Nitro musks are sweet and a little dusty. Polycyclics are sweet and floral, tied to the smell of laundry. Macrocyclics are soft and powdery. Alicyclics are bright with a fruity edge. Ambrette is warm, the closest of all to animal musk. Reading a musk note on a blotter means reading which historical layer it belongs to.
The Lê Mai White Musk perfume belongs to this line: a clean musk note built from molecules, with no material from the musk deer. The move from animal to molecule was not only a matter of ethics. The chemistry of Ružička and those who came after him made it possible, one ring at a time.

The warm smell of skin is now a question of chemistry.
