Hai người, một que thử

Các nhà hóa học của IFF tìm ra Galaxolide năm 1965. Phân tử này rẻ, bền trong môi trường kiềm, mùi sạch, nên nhanh chóng trở thành xạ hương tổng hợp được dùng nhiều nhất thế giới, trong nước hoa, xà bông, nước xả vải và bột giặt. Mùi của nó được tả là xạ sạch, ngọt, có nét hoa và gỗ, mùi của vải vừa giặt.

Một chất phổ biến đến vậy có lẽ ai cũng ngửi thấy. Nhưng không. Galaxolide nằm trong nhóm những phân tử mà một số người không ngửi được, dù khứu giác của họ với mọi mùi khác đều bình thường. Họ không bị cảm lạnh, không mỏi mũi, không có vấn đề gì với hoa hồng hay chanh. Chỉ riêng chất này là không có.

Hiện tượng ấy có tên riêng, và nó giải thích nhiều cuộc trò chuyện khó hiểu quanh một chai nước hoa: người khen mùi xạ dễ chịu, người bảo chai nước hoa gần như không có mùi, người thấy nó mạnh quá. Ba người, một chai, ba mô tả.

Anosmia đặc hiệu là gì

Anosmia là mất khứu giác. Anosmia đặc hiệu là một điều hẹp hơn nhiều: không ngửi được một chất cụ thể trong khi khứu giác bình thường với các chất khác. Theo tài liệu hóa học hương, hiện tượng này khá phổ biến với một số xạ hương tổng hợp và với androstenone.

Anosmia đặc hiệu khác với mỏi khứu giác. Mỏi khứu giác xuất hiện sau khi ngửi lâu một mùi và mất đi khi rời khỏi mùi đó; bài Mười phút sau thì không còn ngửi thấy mô tả cơ chế ấy. Anosmia đặc hiệu có từ trước lần ngửi đầu tiên. Người không ngửi được Galaxolide sẽ không ngửi được nó dù que thử mới hay cũ, dù mũi nghỉ hay không.

Ranh giới cũng không phải lúc nào cũng tuyệt đối. Với androstenone, có những người lúc đầu không phát hiện được chất này nhưng học được cách ngửi nó sau khi tiếp xúc lặp lại. Ngưỡng phát hiện androstenone được ghi nhận xuống tới cỡ phần tỉ ở những người nhạy nhất, nghĩa là khoảng cách giữa người nhạy và người không nhạy có thể rất lớn.

Vì vậy anosmia đặc hiệu nên được hiểu như một dải hơn là một công tắc. Ở một đầu là người ngửi rõ ở nồng độ rất thấp. Ở đầu kia là người không ngửi thấy ở bất kỳ nồng độ nào thử được. Phần lớn nằm đâu đó ở giữa.

Gen thụ thể và biến thể

Ở người, khoảng 400 gene thụ thể mùi còn hoạt động và khoảng 600 gene khác đã thành gene giả. Biến thể di truyền trong họ gene này rất phong phú và làm thay đổi chức năng thụ thể. Hai người có thể mang hai phiên bản khác nhau của cùng một thụ thể, một phiên bản hoạt động tốt, một phiên bản hoạt động kém.

Năm 2019, Trimmer và cộng sự giải trình tự toàn bộ bộ gene thụ thể mùi của 332 người, rồi so với 276 đặc điểm cảm nhận, gồm cường độ và độ dễ chịu của 68 chất mùi ở hai nồng độ. Biến thể ở một thụ thể đơn lẻ thường gắn với thay đổi trong cảm nhận. Nhóm nghiên cứu kiểm chứng 10 trường hợp bằng thử nghiệm chức năng; trong 8 trường hợp, thụ thể hoạt động kém đi kèm với cảm nhận cường độ yếu hơn.

Năm 2022, Li và cộng sự quét toàn bộ hệ gene ở khoảng 1.000 người Hán, rồi kiểm lại ở một nhóm 364 người đa dạng về nguồn gốc. Họ lặp lại được ba liên hệ đã biết: β ionone với OR5A1, androstenone với OR7D4, cis-3-hexenol với vùng gene quanh OR2J3. Họ cũng thấy các alen mới hơn trong lịch sử tiến hóa có xu hướng làm cảm nhận cường độ yếu đi.

Gene không giải thích tất cả. Trong nghiên cứu năm 2019, gene của một thụ thể cộng với nguồn gốc di truyền, tuổi và giới chỉ giải thích được khoảng 10 đến 20 phần trăm khác biệt cảm nhận ở 15 đặc điểm. Phần còn lại thuộc về kinh nghiệm, cách đo, và những gì khoa học chưa tách ra được.

Musk, Iso E Super, androstenone

Androstenone là trường hợp được hiểu rõ nhất. Năm 2007, Keller và cộng sự cho thấy thụ thể OR7D4 phản ứng chọn lọc với androstenone. Người mang biến thể hoạt động kém của thụ thể này, như một nhóm, kém nhạy hơn với chất ấy và thấy nó ít khó chịu hơn. Cùng một phân tử được người này tả là nước tiểu cũ, người khác tả là ngọt hay giống vani, người khác không thấy gì.

Với musk, bức tranh có nhiều thụ thể hơn. OR5AN1 được xem là một thụ thể musk ở người; phản ứng của nó với nhiều loại musk trong ống nghiệm khớp với độ nhạy cảm nhận. Năm 2023, Sato Akuhara và cộng sự ghi nhận người mang hai bản của một biến thể nhạy hơn có ngưỡng phát hiện muscone thấp hơn và thấy các musk vòng lớn mạnh hơn. Riêng Galaxolide, nghiên cứu năm 2022 của Li và cộng sự tìm thấy một thụ thể khác, OR4D6, và đây là thụ thể người đầu tiên được chứng minh gây anosmia đặc hiệu với một hợp chất musk.

Iso E Super là một phân tử gỗ mịn, khô, rất trong, do IFF phát triển đầu thập niên 1970. Thư viện ghi rằng nhiều người nhận biết nó yếu, dù nó làm các mùi xung quanh tỏa rộng hơn. Trong các nguồn đã đối chiếu cho bài này, chưa tìm thấy nghiên cứu gene thụ thể nào giải thích riêng cho Iso E Super; phần này vì thế chỉ dừng ở quan sát.

Ba trường hợp cho ba mức hiểu biết khác nhau: một thụ thể đã xác định cho androstenone, hai thụ thể cho hai nhóm musk, và một câu hỏi còn mở cho Iso E Super.

Vì sao nước hoa dùng nhiều musk

Xạ hương tổng hợp không phải một chất mà là nhiều họ phân tử. Musk vòng lớn như muscone, habanolide, ethylene brassylate. Musk đa vòng như Galaxolide, tonalide. Musk nitro, nhiều chất trong đó nay đã bị IFRA cấm. Mỗi họ có hình dạng khác, và như các nghiên cứu ở trên cho thấy, có thể được những thụ thể khác nhau nhận ra.

Từ đó có một hệ quả logic. Nếu musk vòng lớn được nhận chủ yếu qua một thụ thể và Galaxolide qua một thụ thể khác, thì một người không ngửi được một loại vẫn có thể ngửi được loại kia. Dùng nhiều họ musk trong cùng một công thức làm giảm khả năng phần đáy biến mất hoàn toàn với một người đeo nào đó. Đây là suy luận từ dữ liệu thụ thể; các nguồn khoa học của bài không nêu nó như lý do chính thức của các nhà pha hương.

Nhiều công thức hiện đại còn đặt musk cạnh ambroxide và Iso E Super. Ambroxide cho mùi hổ phách khô, sạch, có nét gỗ và khoáng, như da ấm sau nắng, rất tỏa và rất bền. Cặp ambroxide với Iso E Super là nền của nhiều nước hoa gỗ hổ phách tối giản. Một nền gồm nhiều phân tử khác loại như vậy khó bị một mũi bỏ sót toàn bộ.

Trong dữ liệu thư viện, những mùi xạ được tả là sạch như vải giặt gắn với các phân tử như Galaxolide và habanolide. Galaxolide cũng là musk đầu tiên có một thụ thể gây anosmia đặc hiệu được xác định, nên ở nhóm mùi sạch này, khác biệt giữa các mũi có cơ sở rõ nhất.

Mỗi mũi một bộ lọc

Khoảng 400 loại thụ thể, mỗi loại có thể có nhiều phiên bản, tạo ra một tổ hợp gần như riêng cho từng người. Một que thử không có một mùi duy nhất; nó có bao nhiêu mùi là tùy bao nhiêu mũi ngửi nó.

Điều này thay đổi cách đọc một lời mô tả nước hoa. Khi một chai được tả là xạ mềm, gỗ trong, đó là mô tả của một hay vài người, qua bộ lọc của họ. Một người khác có thể đọc cùng chai ấy thành gỗ khô, hoặc gần như không mùi. Không ai trong số họ sai.

Nó cũng thay đổi cách thử. Nếu một phần của chai không hiện ra, phần đó có thể nằm ngoài bộ lọc của người ngửi, chứ không phải vắng mặt trong chai. Two People, One Blotter

IFF chemists discovered Galaxolide in 1965. The molecule is cheap, stable in alkaline media and clean-smelling, so it quickly became the most used synthetic musk in the world, in perfume, soap, fabric softener and detergent. Its smell is described as clean, sweet musk with floral and woody touches, the smell of freshly laundered cloth.

A material that common, one might think, is smelled by everyone. It is not. Galaxolide belongs to the group of molecules that some people cannot smell, even though their sense of smell is normal for everything else. They do not have a cold, their nose is not fatigued, they have no difficulty with rose or lemon. Only this substance is missing.

The phenomenon has its own name, and it explains many puzzling conversations around a bottle of perfume: one person praises a pleasant musk, another says the perfume barely smells, a third finds it too strong. Three people, one bottle, three descriptions.

What Specific Anosmia Is

Anosmia is loss of the sense of smell. Specific anosmia is much narrower: the inability to smell one particular substance while smelling others normally. According to the fragrance chemistry literature, it is fairly common with some synthetic musks and with androstenone.

Specific anosmia differs from olfactory fatigue. Fatigue appears after smelling one odour for a long time and goes once the odour is left behind; the post Ten Minutes Later, Gone describes that mechanism. Specific anosmia is there before the first sniff. Someone who cannot smell Galaxolide will not smell it whether the blotter is fresh or old, whether the nose is rested or not.

The boundary is not always absolute either. With androstenone, some people who at first cannot detect it learn to smell it after repeated exposure. Detection thresholds for androstenone have been reported down to around parts per billion in the most sensitive people, so the gap between sensitive and insensitive noses can be very wide.

Specific anosmia is therefore better understood as a range than a switch. At one end are people who smell a substance clearly at very low concentration. At the other are people who smell nothing at any concentration tested. Most sit somewhere in between.

Receptor Genes and Their Variants

In humans about 400 odour receptor genes remain functional and about 600 others have become pseudogenes. Genetic variation in this family is abundant and alters receptor function. Two people may carry two versions of the same receptor, one working well, the other poorly.

In 2019 Trimmer and colleagues sequenced the full odour receptor repertoire of 332 people and compared it with 276 perceptual traits, including the intensity and pleasantness of 68 odorants at two concentrations. Variation in a single receptor was frequently associated with changes in perception. The team validated 10 cases with a functional assay; in 8 of them, reduced receptor function went with weaker perceived intensity.

In 2022 Li and colleagues scanned the whole genome in about 1,000 Han Chinese and checked the results in an ethnically diverse group of 364. They replicated three known links: β ionone with OR5A1, androstenone with OR7D4, cis-3-hexenol with the gene region around OR2J3. They also found that the evolutionarily derived alleles tended to reduce perceived intensity.

Genes do not explain everything. In the 2019 study, one receptor's genotype together with genetic ancestry, age and gender explained only about 10 to 20 percent of the perceptual variation in 15 traits. The rest belongs to experience, to how things are measured, and to what science has not yet separated out.

Musk, Iso E Super, Androstenone

Androstenone is the best understood case. In 2007 Keller and colleagues showed that the receptor OR7D4 responds selectively to androstenone. People carrying a poorly functioning variant of that receptor were, as a group, less sensitive to it and found it less unpleasant. The same molecule is described by one person as stale urine, by another as sweet or vanilla-like, and by another as nothing.

With musk the picture involves more receptors. OR5AN1 is regarded as a human musk receptor; its responses to many musks in vitro match perceived sensitivity. In 2023 Sato Akuhara and colleagues reported that people carrying two copies of a more sensitive variant had a lower detection threshold for muscone and found macrocyclic musks more intense. For Galaxolide, the 2022 study by Li and colleagues found a different receptor, OR4D6, the first human receptor shown to drive specific anosmia to a musk compound.

Iso E Super is a smooth, dry, very transparent woody molecule developed by IFF in the early 1970s. The library notes that many people perceive it only faintly, even though it makes surrounding notes radiate. Among the sources checked for this post, no receptor gene study explains Iso E Super specifically; this part therefore stays at observation.

The three cases show three levels of understanding: an identified receptor for androstenone, two receptors for two groups of musk, and an open question for Iso E Super.

Why Perfumes Use Several Musks

Synthetic musk is not one substance but several families of molecules. Macrocyclic musks such as muscone, habanolide, ethylene brassylate. Polycyclic musks such as Galaxolide and tonalide. Nitro musks, several of them now prohibited by IFRA. Each family has a different shape and, as the studies above show, may be recognised by different receptors.

A logical consequence follows. If macrocyclic musks are detected mainly through one receptor and Galaxolide through another, a person who cannot smell one type may still smell the other. Using several musk families in one formula lowers the chance that the base disappears entirely for a given wearer. This is an inference from receptor data; the scientific sources for this post do not state it as perfumers' official reasoning.

Many modern formulas also place musks beside ambroxide and Iso E Super. Ambroxide smells of dry, clean amber with woody and mineral facets, like warm skin after sun, very diffusive and long-lasting. The pairing of ambroxide with Iso E Super underlies many minimalist woody amber perfumes. A base of several unlike molecules is hard for any single nose to miss completely.

In the library data, musk smells described as clean like laundered cloth are tied to molecules such as Galaxolide and habanolide. Galaxolide is also the first musk for which a receptor driving specific anosmia has been identified, so within this clean group the differences between noses have the clearest basis.

Every Nose a Filter

About 400 receptor types, each potentially in several versions, form a combination close to unique for each person. A blotter does not have one smell; it has as many smells as there are noses smelling it.

This changes how a perfume description reads. When a bottle is described as soft musk and transparent wood, that is the description of one or a few people, through their filters. Someone else may read the same bottle as dry wood, or as almost no smell at all. None of them is wrong.

It changes how to test, too. If part of a bottle does not appear, it may lie outside the filter of the person smelling, rather than being absent from the bottle. Differences between noses are not a defect to be corrected. They are the normal structure of human smell, written into genes.

A bottle of perfume does not have one smell.

It has a list of molecules, and each nose reads part of that list.

The rest is still there, for someone else.
